Biochem/physiol Actions
Exhibits anti-inflammatory activities. Small molecule inhibitor of a novel E3 or IκBα ubiquitin ligase distinct from SCFβTRCP (Skp1-cullin-F box conplex). Prevents IκBα degradation in cell culture (IC50 2.3 ± 0.09 μM). Blocks expression of NF-κB dependent cytokines: TNFα, IL-1β, and IL-6 in both cell culture (IC50 600-700 nM) and in vivo in animal models (NF-KB gene activation requires degradation of IKBα upon phosphorylation by IKK followed by ubiquitination).
